Background: LXR alpha/NR1H3

LXR (liver X receptors - LXR-alpha and LXR-beta, encoded by NR1H3 and NR1H2, respectively) belongs to nuclear hormone receptor superfamily and are ligand-dependent nuclear transcription factors which are activated by oxysterols. LXR beta is ubiquitously expressed whereas LXR alpha is expressed mainly in liver, intestine, adipose tissue and macrophages. LXR-alpha and LXR-beta shares 78% similarity within their ligand-binding domains and form obligate heterodimers with retinoic X receptor (RXR), bind specific response elements within promoters of target genes ABCA1, ABCG1, SREBF1, APOE etc., and regulate their expression. In the unliganded state, LXRs binds with corepressors such as N-CoR which displaces upon ligand binding. Oxysterols (oxidized cholesterol derivatives) are their natural ligands whereas endogenous ligands includes 22(R)-hydroxycholesterol (22-HC), 24(S)-hydroxycholesterol (24(S)-HC), 24,25-EC, 27-hydroxycholesterol etc. LXRs control the major metabolic pathways such as cholesterol homeostasis, lipogenesis, inflammation, toxicity proliferation cell survival/death and different aspacts of acquired/innate immunity.

LXR alpha/NR1H3 Antibody:
Western Blot Protocol

1. Perform SDS-PAGE (4-12%) on samples to be analyzed, loading 50 ug of total protein per lane.

2. Transfer proteins to Nitrocellulose according to the instructions provided by the manufacturer of the transfer apparatus.

3. Rinse membrane with dH2O and then stain the blot using ponceau S for 1-2 minutes to access the transfer of proteins onto the nitrocellulose membrane. Rinse the blot in water to remove excess stain and mark the lane locations and locations of molecular weight markers using a pencil.

4. Rinse the blot in TBS for approximately 5 minutes.

5. Block the membrane using 5% non-fat dry milk + 1% BSA in TBS for 2 hours at room temperature.

6. Rinse the membrane in dH2O and then wash the membrane in wash buffer [TBS + 0.1% Tween] 3 times for 10 minutes each.

7. Dilute the rabbit anti-LXR primary antibody (NB 400-157) in blocking buffer and incubate 30 minutes at room temperature on nutator and then overnight at 4 degrees Celsius.

8. Rinse the membrane in dH2O and then wash the membrane in wash buffer [TBS + 0.1% Tween] 3 times for 10 minutes each.

9. Apply the diluted rabbit-IgG HRP-conjugated secondary antibody in blocking buffer (as per manufacturer's instructions) and incubate 1 hour at room temperature.

10. Wash the blot in wash buffer [TBS + 0.1% Tween] 3 times for 10 minutes each (this step can be repeated as required to reduce background).

11. Apply the detection reagent of choice in accordance with the manufacturer's instructions (Pierce's ECL).

Note: Tween-20 can be added to the blocking or antibody dilution buffer at a final concentration of 0.05-0.2%, provided it does not interfere with antibody-antigen binding.

  • Q: I have a question regarding the amounts of cell extract to use with this product. You show gels from liver and skeletal muscle to demonstrate quality of the antibody, but the amounts of tissue extract used on these gels is not indicated. We are just beginning to work with this antibody, and this information would be helpful.

    A: For our product NB400-157, the blot image showing the human liver lysate was run with 25ug of human liver lysate loaded into the gel. For the other image with the human skeletal muscle lysate, this blot was performed on the ProteinSimple Wes platform. This system utilizes a capillary system to separate the proteins through instead of a traditional gel. This system therefore requires much less material. For this experiment, our lab used 5ul of lysate at 0.2mg/ml.
