Background: Lysine Hydroxylase 2/PLOD2

The protein encoded by this gene is a membrane-bound homodimeric enzyme that is localized to the cisternae of the rough endoplasmic reticulum. The enzyme (cofactors iron and ascorbate) catalyzes the hydroxylation of lysyl residues in collagen-like peptides. The resultant hydroxylysyl groups are attachment sites for carbohydrates in collagen and thus are critical for the stability of intermolecular crosslinks. Some patients with Ehlers-Danlos syndrome type VIB have deficiencies in lysyl hydroxylase activity. Mutations in the coding region of this gene are associated with Bruck syndrome. Alternative splicing results in multiple transcript variants encoding different isoforms. [provided by RefSeq]

  • Q: I have been looking for anti PLOD2 antibody to perform IHC on human lung cancer tissue. I have tried some antibodies from different companies, but it did not work for us well. I found this antibody cat# H00005352-B01P at Novus Biologicals website. On the data sheet you reported it for ICC and IF. I have been using your different antibodies in the past and had some satisfying results. May I ask you, please, if you had tried IHC on human tissue and it did not work or you have not tried it at all?

    H00005352-B01P is a product we distribute for a Taiwanese company called Abnova. We therefore do not do any in-house testing or development of their products. Abnova has validated this PLOD2 antibody for use in western via detection of the recombinant protein and overexpression transfected lysates. They have also validated this antibody in ICC with HeLa cells. They have not yet tested this antibody in tissue samples. See their datasheet here for all information we have regarding this product. Since this is not our product, we unfortunately cannot offer free samples. If you did want to try this product in an IHC experiment, we could offer you our Innovator's Reward Program.
