Western Blot: MDM2/HDM2 [p Ser166] Antibody (JE64-20) [NBP3-32777] - Western blot analysis of MDM2/HDM2 on different lysates with Rabbit anti-MDM2/HDM2 antibody (NBP3-32777) at 1/1,000 dilution.

Lane 1: A549 cell lysate
Lane 2: A549 treated with pp for 1 hour cell lysate

Observed band size: 110/140 kDa

Exposure time: 57 seconds;

4-20% SDS-PAGE gel.

Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (NBP3-32777) at 1/1,000 dilution was used in 5% NFDM/TBST at 4C overnight. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1/50,000 dilution was used for 1 hour at room temperature.

Background: MDM2/HDM2

MDM2 is a target gene of the transcription factor tumor protein p53. The encoded protein is a nuclear phosphoprotein that binds and inhibits transactivation by tumor protein p53, as part of an autoregulatory negative feedback loop. Overexpression of this gene can result in excessive inactivation of tumor protein p53, diminishing its tumor suppressor function. This protein has E3 ubiquitin ligase activity, which targets tumor protein p53 for proteasomal degradation. This protein also affects the cell cycle, apoptosis, and tumorigenesis through interactions with other proteins, including retinoblastoma 1 and ribosomal protein L5. More than 40 different alternatively spliced transcript variants have been isolated from both tumor and normal tissues.
