Recombinant fragment covering tandem repeats of human MUC2 protein (exact sequence is proprietary) (Uniprot: Q02817 )
Reactivity Notes
Does not react with Rat.
Localization
Cytoplasmic and cell surface
Specificity
Recognizes a single glycoprotein of 520kDa, identified as mucin 2 (MUC2). This monoclonal antibody shows no cross-reaction with human milk fat globule membranes, MUC1, or MUC3. Mucins are high molecular weight glycoproteins, which constitute the major component of the mucus layer that protects the gastric epithelium.MUC2 is specifically expressed in goblet cells of the small intestine in about 65% of colonic carcinomas and about 40% of gastric carcinomas. MUC2 is rarely expressed outside of the GI tract with the exceptions of mucinous carcinoma of breast and clear cell-type carcinomas of the ovary.

Scientific Data Images for MUC2 Antibody (MLP/842)
Immunofluorescent Staining of Adult Stem Cell-derived Human Descending Colon Organoids.
Adult stem cells isolated from human descending colon were cultured following the steps detailed in the human intestinal organoid culture protocol. (A) The organoids were fixed and stained with a Mouse Anti-Human MUC2 Monoclonal Antibody (Novus Biologicals; Catalog # NBP2-44431; green) to visualize intestinal goblet cells and counterstained with a Goat Anti-Human/Mouse E-Cadherin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF748; red) and DAPI (Catalog # 5748; blue). The image shown was taken at 10x magnification. (B) The organoids were fixed and stained with a Mouse Anti-Human Chromogranin A Monoclonal Antibody (Catalog # MAB90981; green) to visualize enteroendocrine cells and counterstained with a Goat Anti-Human/Mouse E-Cadherin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF748; red) and DAPI (Catalog # 5748; blue). The image shown was taken at 20x magnification.Immunohistochemistry-Paraffin: MUC2 Antibody (MLP/842) [NBP2-44431]

Immunohistochemistry-Paraffin: MUC2 Antibody (MLP/842) [NBP2-44431] - Human Colon Carcinoma stained with MUC2 Monoclonal Antibody (MLP/842).Applications for MUC2 Antibody (MLP/842)
Application
Recommended Usage
Flow Cytometry
0.5-1 ug/million cells
Immunocytochemistry/ Immunofluorescence
1-2 ug/ml
Immunofluorescence
1 - 2 ug/ml
Immunohistochemistry-Paraffin
1-2 ug/ml
Application Notes
Immunohistochemistry (Formalin-fixed): 1-2ug/ml for 30 minutes at RT. Staining of formalin-fixed tissues requires heating tissue sections in 10mM Tris with 1mM EDTA, pH 9.0, for 45 min at 95C followed by cooling at RT for 20 minutes.

Background: MUC2
Changes or perturbations to MUC2 expression has been associated with a number of disease pathologies (1-4, 6). Specifically, altered MUC2 composition has been indicated in colorectal cancer, inflammatory bowel diseases (IBD) including ulcerative colitis and Chron's disease, and chronic obstructive pulmonary disease (COPD) (1-4, 6). In general, decreased or reduced MUC2 expression is associated with colorectal cancer disease progression and development of IBD (1-4, 6). Additionally, studies have found that upon intestinal infection due to parasites or bacteria MUC2 expression is increased, as is overall mucus secretion (3, 6).
