Flow Cytometry: Neutrophil Elastase/ELA2 Antibody (39A) [Alexa Fluor® 488] [NB100-64392AF488] - Leukocytes were isolated from BAL fluid of a CF patient and stained for analysis by flow cytometry. Macrophages were found to have scavenged substantial neutrophil elastase from the airways while minimal was observed on the surface of T cells from the same patient as expected. This antibody works well for staining multiple cell types and obtains reliable fluorescence in the FITC channel. When used for single-stained beads compensation controls, consistent signal is obtained provided that the cytometer is accurately calibrated. Background: Neutrophil Elastase/ELA2

The ELANE gene (commonly known as neutrophil elastase) encodes a 267 amino acid long, 28 kDA neutrophil elastase protein that functions in regulation of natural killer cells, monocytes, and graulocytes. ELANE is involved in activation of proMMP8, cell adhesion cell-matrix glycoconjugates, selected targets of C/EBPbeta, degradation of the extracellular matrix and collagen, and amb2 integrin signaling. It interacts with various genes GZMB, LRP1, SERPINF2, SERPING1, and SERPINA1. Elane has been linked to cystic fibrosis, asthma, pneumonia, alzheimer's disease, neutropenia, cutis laxia, leg ulcers, wegener's granulomatosis, bronchitis, adult respiratory distress syndrome, and vascular diseases.

    I used this antibody to stain for surface-bound neutrophil elastase. This enzyme is released from neutrophils following exocytosis of the primary granules and can be captured on the surface of neutrophils and macrophages, but less so on lymphocytes. I purified cells from the BAL of cystic fibrosis patients and stained for analysis by flow cytometry. Macrophages were found to have large amounts of surface-bound elastase, while it is minimal on the T cells from the same patient as expected. This antibody works well for staining multiple cell types and obtains reliable fluorescence in the FITC channel. When used for single-stained beads compensation controls, consistent signal is obtained provided that the cytometer is accurately calibrated. One thing to be aware of is that different lots of this antibody have slightly different concentrations.
