Western Blot: Otubain-1 Antibody [NBP1-49934] - Detection of Human and Mouse OTUB1 by Western Blot (h&m) and Immunoprecipitation (h). Samples: Whole cell lysate from HeLa (5, 15 and 50 mcg for WB; 1 mg for IP, 20% of IP loaded), 293T (T; 50 mcg) and mouse NIH3T3 (M; 50 mcg) cells. Antibodies: Affinity purified rabbit anti-OTUB1 antibody used for WB at 0.04 mcg/ml (A) and 0.4 mcg/ml (B) and used for IP at 3 mcg/mg lysate. OTUB1 was also immunoprecipitated by rabbit anti-OTUB1 antibody which recognizes a downstream epitope. For blotting immunoprecipitated OTUB1 was used. Detection: Chemiluminescence with exposure times of 30 seconds (A) and 10 seconds (B).

OTUB1 inhibits breast cancer growth in vivo. (A) Control, OTUB1−/− and OTUB1−/− + CCN6 4T1 cells were injected into nude mice and tumour volume was measured (n = 6 for all groups) (mean +/- SEM, NC vs. OTUB1−/−: *p <.05, **p <.01; NC vs. OTUB1−/− + CCN6: #p <.05, ##p <.01). On day 9 after tumour inoculation, mice were sacrificed and tumours were collected. Tumour (B) size and (C) weight were measured (n = 6 for all groups) (mean +/- SEM, *p <.05, **p <.01). (D) Whole‐cell lysates of tumour samples were analysed by Western blot for OTUB1, CCN6 and GAPDH. (E) Tumour samples were stained for Ki‐67 and immunoreactivity was detected by 3,3′‐diaminobenzidine chromogen (brown). Sections were counterstained with haematoxylin (blue) (scale bar = 100 μm). Background: Otubain-1

The product of this gene is a member of the OTU (ovarian tumor) superfamily of predicted cysteine proteases. The encoded protein is a highly specific ubiquitin iso-peptidase, and cleaves ubiquitin from branched poly-ubiquitin chains but not from ubiquitinated substrates. It interacts with another ubiquitin protease and an E3 ubiquitin ligase that inhibits cytokine gene transcription in the immune system. It is proposed to function in specific ubiquitin-dependent pathways, possibly by providing an editing function for polyubiquitin chain growth. Alternative splicing results in multiple transcript variants. [provided by RefSeq]
