IHC: NB110-40461 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity..

Background: PDE6A

This gene encodes the cyclic-GMP (cGMP)-specific phosphodiesterase 6A alpha subunit, expressed in cells of the retinal rod outer segment. The phosphodiesterase 6 holoenzyme is a heterotrimer composed of an alpha, beta, and two gamma subunits. cGMP is an important regulator of rod cell membrane current, and its dynamic concentration is established by phosphodiesterase 6A cGMP hydrolysis and guanylate cyclase cGMP synthesis. The protein is a subunit of a key phototransduction enzyme and participates in processes of transmission and amplification of the visual signal. Mutations in this gene have been identified as one cause of autosomal recessive retinitis pigmentosa.

  • Q: Could you by any chance provide more information on the antibody in immunohistochemistry? The picture shown in human is not clear and does not look specific. We are looking to try this out in frozen sections in dog. If you cannot provide this information, would it be possible to obtain a small aliquot for testing before purchasing your antibody? We have tried the PDE6B antibody and it works very cleanly in dog retina, and wish to add to this for a publication, to get PDE6A staining.

    A: PDE6A in dogs shares 95% homology so this antibody is predicted to cross-react with canine samples, however, Novus is not able to offer free samples at this time.
