Immunohistochemistry-Paraffin: Podoplanin Antibody (NZ-1.2) [NBP2-03952] - Podoplanin (NZ-1.2) was detected in immersion fixed paraffin-embedded sections of human appendix using Rat Anti-Human Podoplanin Monoclonal Antibody (Catalog # NBP2-03952) at 0.5 ug/mL for 1 hour at room temperature followed by incubation with the Anti-Rat IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC005). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to the cell surface.

Background: Podoplanin

Podoplanin is a mucin type-1 glycoprotein of 40-43kd also know as T1A, gp38 or Aggrus. Podoplanin is expressed in many tumors and many normal cells, especially lymphatic epithelial cells and follicular DCs. It appears to serve as a ligand for CLEC-2. Podoplanin expression is positively correlated with tumors expressing greater invasive and metastatic potential. Podoplanin is directly involved in cell migration, aids metastases formation and tumor cell invasion of tissue (Schacht, 2005, Raica, 2008, Martin-Villar, 2010). It has also been determined that podoplanin expression is induced through Src activation of Cas (Shen, 2010). Recently there are indications of podoplanin presence on Th17 cells (Bettelli, 2011). Initial studies below indicate the presence of podoplanin on a small but distinct population of CD4+ cells from human PBMC.
