Immunocytochemistry/ Immunofluorescence: PSENEN/PEN2 Antibody (6J5O4) [NBP3-16676] - Confocal imaging of NIH/3T3 cells using PSENEN/PEN2 Rabbit mAb followed by a further incubation with Cy3 Goat Anti-Rabbit IgG (H+L). The cells were counterstained with alpha-Tubulin Mouse mAb followed by incubation with ABflo(R) 488-conjugated Goat Anti-Mouse IgG (H+L) Ab (Green). DAPI was used for nuclear staining (Blue). Objective: 100x.

Background: PSENEN

Presenilin (PS1/PS2) complexes are responsible for the intramembrane gamma-secretase cleavage of the beta-amyloid precursor protein. Presenilin enhancer 2 (PEN2) is an essential cofactor involved in physically interacting with PS1 and is necessary for gamma-secretase activity. Endogenous PEN2 preferentially interacts with PS1 holoprotein. Down-regulation of PEN2 expression abolishes the endoproteolysis of PS1 and overexpression promotes the production of PS1 fragments.
