Flow Cytometry: RHD Antibody (D7C2) - Chimeric - Azide and BSA Free [NBP3-09042] - Paraformaldehyde fixed K562 cells were stained with anti-unknown specificity antibody (isotype control, black line) or NBP3-09042 (blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.

This chimeric rabbit antibody was made using the variable domain sequences of the original Human IgM format, for improved compatibility with existing reagents, assays and techniques.

FC confirmed the binding of D7C2 to Rh D+ red cells in artifical mixtures (Edelman et al, 1997). The ADCC of D7C2 showed a percentage of 93% lysis (Edelman et al, 1997). D7C2 has been expressed recombinantly using a baculovirus-insect cell expression system (Edelman et al, 1997).

Background: RHD

The Rh blood group system is the second most clinically significant of the blood groups, second only to ABO. It is also the most polymorphic of the blood groups, with variations due to deletions, gene conversions, and missense mutations. The classification of Rh positive and Rh negative individuals is determined by the presence or absence of the highly immunogenic RhD protein on the surface of erythrocytes. Alternative splicing of the gene results in four transcript variants encoding four different isoforms.
