Western Blot: Roquin Antibody [NB100-656] - Samples: A) Whole cell lysate from HeLa (5, 15 and 50 ug) and 293T (T, 50 ug) cells. B) Whole cell lysate (1 mg/IP; 1/4 of IP loaded) from HeLa cells. Antibodies: Affinity purified rabbit anti-Roquin antibody NB100-656 used at 0.04 ug/ml for WB (A and B) and at 3 ug/mg lysate for IP (B). Roquin was also immunoprecipitated using rabbit anti-Roquin antibody NB100-655 at 3 ug/mg lysate. Detection: Chemiluminescence with an exposure time of 30 seconds.

Background: Roquin

Roquin is a member of the RING-type E3 ubiquitin ligase protein family. It was recently found to contribute to autoimmune diseases such as type 1 diabetes and lupus. Research has found that a mutation in roquin causes T-cells to attack their own tissues. Roquin stops T-cells from displaying a stimulatory receptor, ICOS, that may cause the cells to attack normal body tissues. (1)
