Description

This antibody is highly specific and optimized for use in Immunohistochemistry.

It has been validated on TMAs covering 76 normal tissues and numerous cancer tissues.

Positive Control
Postive control:Colon: A moderate to strong staining of the axons of the Auerbach's and Meissner's plexus should be seen while the endocrine cells of the mucosa and the peripheral nerves in the mucosa must show an at least a weak to moderate staining. Pancreas: A strong staining should be seen in islets of Langerhans and in peripheral nerve fibres located between the exocrine cells.

Negative Control
Negitve control:Colon: All non-endocrine epithelial cells and lymphocytes should not show any staining.

Application Notes
Manual Protocol: Freshly cut sections should be used (less than 10 days between cutting and staining). Heat-induced antigen retrieval for 5 minutes in an autoclave at 121C in pH 7,8 Target Retrieval Solution buffer. Apply the antibody at a dilution of 1:150 at 37C for 60 minutes. Visualization of bound antibody by the EnVision Kit (Dako, Agilent) according to the manufacturer's directions.

Background: Synaptophysin

Synaptophysin (Syp I), also called the major synaptic vesicle protein p38, is an integral membrane glycoprotein found in the synaptic vesicles of brain, neuron and neuroendocrine cells. Synaptophysin is one of the most abundant membrane proteins, comprising approximately 7% of the total protein in small synaptic vesicles. In mature nerve terminals it forms a complex with the vesicular membrane protein synaptobrevin, which appears to modulate synaptobrevin's interaction with the plasma membrane-associated proteins syntaxin and SNAP25 to form the SNARE complex as a prerequisite for membrane fusion. Synaptophysin may also be used as a neuroendocrine tumor marker in both neuroscience and cancer research.
