Specificity

This product recognises testosterone, a steroid hormone and most potent of the naturally occurring androgens. Testosterone promotes the development and maintenance of male secondary sex characteristics and is produced primarily in the testes, but is also produced in small amounts by the adrenal glands, ovaries and placenta. Synthetic derivatives of testosterone are used therapeutically in the treatment of breast cancer and for hormone replacement therapy. NB100-62943 also recognises 5-a-Dihydrotestosterone (1%) and Androstenedione (1%).

Background: Testosterone

Testosterone is the principal androgen, or male sex hormone. One of the group of compounds known as anabolic steroids, testosterone is secreted by the testes but is also synthesized in small quantities in the ovaries, cortices of the adrenal glands, and placenta, usually from cholesterol. Testosterone is necessary in the fetus for the development of male external genitalia. Testosterone also stimulates protein synthesis and accounts for the greater muscular development of the male. For many years, synthetic steroids similar to testosterone have been used by athletes with the goal of improving performance, although recent medical research has shown that these drugs may have a wide range of harmful side effects.

    Does R&D Systems® Testosterone Parameter Assay Kit, Catalog # KGE010, detect free or total testosterone?

    A:

    'Free' testosterone is typically defined as testosterone which is bioavailable, and not bound to sex hormone binding globulin (SHBG). Interference testing performed by spiking SHBG into testosterone-containing samples, demonstrates interference in testosterone detection with Catalog # KGE010, when the concentration of SHBG is greater than 100 ng/ml. (For further details on interference testing, please refer to the "Specificity" section in the product insert.) 

    Assays for 'total' testosterone measure both 'free' testostrone as well as testosterone bound to SHBG. Since KGE010 does not detect testosterone bound to SHBG, this assay does not measure 'total' testosterone.

  • Q: I would like to ask whether antibody Testosterone # NB100-62943 may be diluted at the beginning of dH2O or Coating Buffer? I would also like to ask whether this antibody are worthy for EIA of non-invasive samples (fecal extract monkey)?

    A: We recommend dilution this antibody in blocking buffer. It should never be diluted in water and if you don't want to use blocking buffer, you should use PBS or TBS as these are also acceptable buffers. Testosterone is the same in all species, so it should work in your samples.
