Background: Tubby

Tubby (TUB) and tubby-like protein-1 (Tulp-1) belongs to tubby protein family that include members such as Tubby, Tulp-1, -2 and -3; Tulps etc. which share a highly conserved C-terminal region of 'tubby domain' with about 260 amino acids.Tubby is the founding member of its family and is expressed in ciliated sensory neurons with cytoplasmic localization. Tubby is associated with PtdIns(4,5)P2 (phosphatidylinositol 4,5-bisphosphate) on the inner leaflet of plasma membrane through its C-terminal domain and receptor-mediated activation of Galphaq (G protein alphaq) releases tubby from the plasma membrane followed by triggering its nuclear translocation. Tubby interacts with rgb-3 and plays a key a role in DAF-16 independent fat regulation as well as in ciliar sensory function which is required for normal sensory behavior such as chemotaxis. Tubby functions in life span control via insulin/IGF-1 pathway and is proposed to be involved in neuronal trafficking also. The naturally occurring tubby mutation in mouse causes retinitis pigmentosa, cochlear degeneration/hearing loss and adult-onset obesity.
