Collagens comprise a large family of insoluble extracellular glycoproteins that are essential components of connective tissues such as tendons, ligaments, cartilage, bone and skin. The mature polypeptides are secreted as coiled, left-handed helices that subsequently assemble into rope-like collagen fibers. Collagen I is a fibril-forming collagen that requires N- and C- terminal processing. Collagen IV is a network forming collagen whose C-terminus forms dimers and N-terminus forms tetramers.

It has been validated on TMAs covering 76 normal tissues and numerous cancer tissues.
Positive Control
Postive control:Colon: An at least moderate collagen IV immunostaining of basement membranes and a strong staining of vessels and the tunica muscularis mucosa should be seen.
Negative Control
Negitve control:Colon: Collagen IV immunostaining must be absent in epithelial cells.

Application Notes
Manual Protocol: Freshly cut sections should be used (less than 10 days between cutting and staining). Heat-induced antigen retrieval for 5 minutes in an autoclave at 121C in pH 7,8 Target Retrieval Solution buffer. Apply the antibody at a dilution of 1:150 at 37C for 60 minutes. Visualization of bound antibody by the EnVision Kit (Dako, Agilent) according to the manufacturer's directions.
