Detection of IL‑9 in Human PBMCs by Flow Cytometry.
Human peripheral blood mononuclear cells (PBMCs) were treated with plate-bound anti-CD3 Monoclonal Antibody (5 μg/ml) and anti-CD28 Monoclonal Antibody (2 μg/ml), Recombinant Human IL-4 (204-IL,10 ng/ml), and anti-Human IFN-gamma Monoclonal Antibody (AF-285-NA, 20 μg/ml) for 5 days and stained with (A) Mouse Anti-Human IL-9 Monoclonal Antibody (Catalog # MAB209) or (B) Mouse IgG2B Isotype Control Antibody (MAB0041) followed by Allophycocyanin-conjugated anti-Mouse IgG Secondary Antibody (F0101B) and Mouse Anti-Human IFN-gamma Phycoerythrin-conjugated Monoclonal Antibody (IC285P). To facilitate intracellular staining, cells were fixed with Flow Cytometry Fixation Buffer (FC004) and permeabilized with Flow Cytometry Permeabilization/Wash Buffer I (FC005).
