Detection of Pax3/Pax7 in Human Skin. Pax3 was detected in immersion fixed paraffin-embedded sections of human skin using Goat Anti-Human Pax3/Pax7 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2457) at 3 µg/ml for 1 hour at room temperature followed by incubation with the Anti-Goat IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC004) or the HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using VisUCyte Antigen Retrieval Reagent-Basic (Catalog # VCTS021). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to the nucleus of melanocytes. Pax3 belongs to the family of paired box transcription factors. Pax family proteins typically contain a paired box domain and a paired-type homeodomain. These transcription factors play critical roles during fetal development. There is 98% homology between human and mouse Pax3.
