Detection of Human TACE/ADAM17 by Western Blot.
Western blot shows lysates of A549 human lung carcinoma cell line and NTera-2 human testicular embryonic carcinoma cell line. PVDF membrane was probed with 2 µg/mL of Mouse Anti-Human TACE/ADAM17 Cytosolic Domain Monoclonal Antibody (Catalog # MAB21291) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for TACE/ADAM17 at approximately 130 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.Detection of TACE/ADAM17 in HeLa Human Cell Line by Flow Cytometry.
HeLa human cervical epithelial carcinoma cell line was stained with Mouse Anti-Human TACE/ADAM17 Cytosolic Domain Monoclonal Antibody (Catalog # MAB21291, filled histogram) or isotype control antibody (Catalog # MAB002, open histogram), followed by Phycoerythrin-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # F0102B). To facilitate intracellular staining, cells were fixed with Flow Cytometry Fixation Buffer (Catalog # FC004) and permeabilized with Flow Cytometry Permeabilization/Wash Buffer I (Catalog # FC005). Western blot shows lysates of HeLa human cervical epithelial carcinoma parental cell line and TACE/ADAM17 knockout HeLa cell line (KO). PVDF membrane was probed with 2 µg/mL of Mouse Anti-Human TACE/ADAM17 Cytosolic Domain Monoclonal Antibody (Catalog # MAB21291) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for TACE/ADAM17 at approximately 140 kDa (as indicated) in the parental HeLa cell line, but is not detectable in knockout HeLa cell line. GAPDH (Catalog # MAB5718) is shown as a loading control.
