Interleukin 27 (IL-27) is a novel heterodimeric cytokine comprised of the IL-12 p35-related protein, p28, and the IL-12 p40-related protein, EBI-3 (Epstein‑Barr virus‑induced gene 3). IL-27 binds TCCR/WSX-1 on naïve CD4+ T cells and induces the expression of a functional IL-12 receptor, making these cells sensitive to IL‑12‑mediated Th1 cell development.

Detection of IL‑27 in LPS-treated RAW 264.7 Mouse Cell Line by Flow Cytometry.
RAW 264.7 mouse monocyte/macrophage cell line was unstimulated (light orange open histogram) or treated with 1 µg/mL LPS for 20 hours, then stained with Rat Anti-Mouse IL-27 p28/IL-30 Monoclonal Antibody (Catalog # MAB1834, dark orange filled histogram) or isotype control (Catalog # MAB0061, blue open histogram), followed by Phycoerythrin-conjugated Anti-Rat IgG F(ab')2Secondary Antibody (Catalog # F0105B). To facilitate intracellular staining, cells were fixed with paraformaldehyde and permeabilized with saponin.
