Background: Pericentrin

Pericentrin is encoded by this gene binds to calmodulin and is expressed in the centrosome. It is an integral component of the pericentriolar material (PCM). The protein contains a series of coiled-coil domains and a highly conserved PCM targeting motif called the PACT domain near its C-terminus. The protein interacts with the microtubule nucleation component gamma-tubulin and is likely important to normal functioning of the centrosomes, cytoskeleton, and cell-cycle progression.

    Smear of -20C methanol-fixed mouse small intestinal epithelial cells stained for ZO-1 (red) and pericentrin (green).
    We tested the antibody on smears (cytology-like preps) of mouse small intestinal epithelial cells fixed with 2% paraformaldehyde/lysine/periodate (PLP; McLean and Nakane. J Histochem Cytochem, 1974) or -20°C methanol. After permeabilizing with 0.1% saponin (PLP-fixed only) and blocking with 5% donkey serum (in PBS), slides were incubated with antibodies at the dilutions indicated below and a rat monoclonal anti-ZO-1 (tight junction protein) antibody (clone R40.76) for 2 hours at room temperature. After washing, species-specific secondary antibodies (cross-adsorbed) produced in goat were applied.
