IFN‑ gamma in Rat Splenocytes.
IFN-gamma was detected in immersion fixed rat splenocytes treated with PMA and calcium ionomycin using Mouse Anti-Rat IFN-gamma Monoclonal Antibody (Catalog # MAB5851) at 25 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (red; Catalog # NL007) and counterstained with DAPI (blue). Specific staining was localized to cytoplasm. Recombinant Rat IFN‑ gamma (Catalog # 585-IF) was serially diluted and captured by Mouse Anti-Rat IFN‑ gamma Monoclonal Antibody (Catalog # MAB5851) coated on a Clear Polystyrene Microplate (Catalog # DY990). Goat Anti-Mouse/Rat IFN‑ gamma Antigen Affinity-purified Polyclonal Antibody (Catalog # AF-585-NA) was biotinylated and incubated with the protein captured on the plate.
