Immunohistochemistry-Paraffin: SARS-CoV-2 ORF8 Antibody [NBP3-11934] - Immunohistochemistry Validation of SARS-CoV-2 (COVID-19) ORF8 in COVID-19 Patient Lung Immunohistochemical analysis of paraffin-embedded COVID-19 patient lung tissue using anti- SARS-CoV-2 ORF8 antibody (NBP3-11934, 0.1 ug/mL). Tissue was fixed with formaldehyde and blocked with 10% serum for 1 h at RT; antigen retrieval was by heat mediation with a citrate buffer (pH6). Samples were incubated with primary antibody overnight at 4C. A goat anti-rabbit IgG H&L (HRP) at 1/250 was used as secondary. Counter stained with Hematoxylin. Strong signal of SARS-COV-2 ORF8 protein was observed in macrophages of COVID-19 patient lung, but not in non-COVID-19 patient lung.ELISA: SARS-CoV-2 ORF8 Antibody - BSA Free [NBP3-11934]

Background: SARS-CoV-2 ORF8
Studies have revealed that SARS-CoV-2 ORF8 has a role in host immune suppression during viral infection as it is an inhibitor of the type I interferon pathway (4, 5). Specifically, ORF8 has been shown to have antagonistic properties on interferon-beta (IFN-beta), the NF-kappabeta-responsive promoter, and the interferon-stimulated response element (ISRE) (4,5). Additionally, ORF8 colocalizes with the ER and lysosomal proteins Calnexin and LAMP1, playing a role in major histocompatibility complex class I (MHC-I) downregulation and degradation (4, 6). These results suggest that inhibiting ORF8 may be a potential approach for improving immune surveillance and more quickly eliminating SARS-CoV-2 infection (4-6).
