Specificity

ACTH (same as Corticotropin) is a 39 amino acid active peptide produced by the anterior pituitary. This monoclonal antibody is specific to Synacthen (aa1-24 of ACTH); does not react with CLIP (aa17-39 of ACTH). POMC (pro-opiomelanocortin or corticotropin-lipotropin) is a 267 amino acid polypeptide hormone precursor that goes through extensive, tissue-specific posttranslational processing by convertases. POMC is cleaved into ten hormone chains named NPP, ACTH, alpha-MSH (Melanocyte Stimulating Hormone), beta-MSH, gamma-MSH, CLIP (corticotropin-like intermediary peptide), Lipotropin-beta, Lipotropin-gamma, beta-endorphin and Met-enkephalin. ACTH is also produced by cells of immune system (T-cells, B-cells, and macrophages) in response to stimuli associated with stress. Anti-ACTH is a useful marker in classification of pituitary tumors and the study of pituitary disease. It reacts with ACTH-producing cells (corticotrophs).It also may react with other tumors (e.g. some small cell carcinomas of the lung) causing paraneoplastic syndromes by secreting ACTH. '

Background: ACTH

ACTH occurs in cells of the anterior pituitary and in neurons in brain. It regulates the corticosteroid production in the adrenal cortex. Beta endorphin and Met enkephalin are endogenous opiates. MSH (melanocyte stimulating hormone) increases the pigmentation of skin by increasing melanin production in melanocytes.

  • Q: The product datasheet indicates that I should store at 4 degrees for short term, and aliquot/store at -20 degrees long term. What constitutes both short and long term?

    A: For storage, any time longer than 1 week, we would consider long term storage. Likewise, short term storage would be anything up to a week. These are just recommendations based on what we have found to be optimal for these products. We also recommend aliquoting and storing material not in use to avoid freeze thaw cycles which can lead to loss of activity of the antibody. The reason we recommend long term storage at -20 is that we have found that these products, when stored at 4C long term, tend to lose their proper function well before the one year guarantee period.
