Product Specifications
Immunogen
The original antibody was generated by immunizing female ICR mice with recombinant human CK20 histidine-tagged fusion protein in Freund's complete adjuvant. (Uniprot# P35900)
Specificity
This antibody binds human cytokeratin 20. Cytokeratins are intermediate filaments found in the intracytoplasmic cytoskeleton of epithelial tissue. There are 19 keratin polypeptides are found in human epithelium. Cytokeratin 20 plays a significant role in maintaining keratin filament organization in intestinal epithelia. When phosphorylated, it plays a role in the secretion of mucin in the small intestine.

Description
This chimeric rabbit antibody was made using the variable domain sequences of the original Mouse IgG2a format for improved compatibility with existing reagents assays and techniques.

Western Blot: Cytokeratin 20 Antibody (10A2) - Chimeric - BSA Free [NBP3-43568] -
Western Blot: Cytokeratin 20 Antibody (10A2) - Chimeric - BSA Free [NBP3-43568] - Human duodenum (A) (0.01µg/ml), ileum (B) (0.001µg/ml), and colon (C) (0.001µg/ml) tissue lysate (35µg protein in RIPA buffer) were resolved on an SDS-PAGE gel, and blots were probed with NBP3-43568 at the mentioned respective concentrations before detection using an anti-rabbit secondary antibody. A primary incubation of 1 hour was used, and protein was detected by chemiluminescence.Immunocytochemistry/ Immunofluorescence: Cytokeratin 20 Antibody (10A2) - Chimeric - BSA Free [NBP3-43568] -
Immunocytochemistry/ Immunofluorescence: Cytokeratin 20 Antibody (10A2) - Chimeric - BSA Free [NBP3-43568] - Immunofluorescence analysis of paraformaldehyde fixed Caco-2 cells, permeabilized with 0.15% Triton, on coverslips stained with NBP3-43568 (1:100 dilution) for 1h followed by Alexa Fluor® 488 secondary antibody (1:1000 dilution), showing cytoplasmic staining. The nuclear stain is DAPI (blue). Panels show, from left-right, top-bottom, NBP3-43568, DAPI, merged channels, and an isotype control. The isotype control was an unknown specificity antibody followed by staining with Alexa Fluor® 488 secondary antibody.Flow Cytometry: Cytokeratin 20 Antibody (10A2) - Chimeric - BSA Free [NBP3-43568] -
Flow Cytometry: Cytokeratin 20 Antibody (10A2) - Chimeric - BSA Free [NBP3-43568] - Paraformaldehyde fixed Caco-2 cells permeabilized with 0.5% Triton were stained with the anti-unknown specificity antibody (isotype control, black line) or NBP3-43568 (blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor® 488 antibody at a dilution of 1:1000, and the cells were analyzed using a FACSCanto flow-cytometer.Applications for Cytokeratin 20 Antibody (10A2) - Chimeric - BSA Free
