Specificity

Epitope of this monoclonal antibody is located in the 8th type III repeat in the cell-binding region of fibronectin. Fibronectins are disulfide-linked, dimeric glycoproteins of ~440kDa. They possess at least four binding sites for collagen, glycosaminoglycans, transglutaminase, and a cell surface receptor. Fibronectins are involved in cell adhesion, tissue organization, and wound healing. Fibronectins are present in basement membranes, interstitial connective tissue matrix, and blood. Cellular fibronectin is widely distributed in the stroma of many malignant tumors. This monoclonal antibody is excellent for staining of formalin-fixed, paraffin-embedded tissues.

Use in Immunocytochemistry/Immunofluorescence reported in scientific literature (PMID:34474855)
Immunohistochemistry (Formalin-fixed): 1-2ug/ml for 30 minutes at RT. Staining of formalin-fixed tissues requires heating tissue sections in 10mM Tris with 1mM EDTA, pH 9.0, for 45 min at 95C followed by cooling at RT for 20 minutes.

Background: Fibronectin

Fibronectin (also known as FN1, or Fibronectin 1) is an extracellular matrix glycoprotein that binds to integrins and has a large role in cell adhesion, migration, differentiation, and growth. It plays a key role in wound healing by helping form blood clots and participates in embryonic development by guiding cell attachment and migration. Fibronectin consists of two monomers linked by disulfide bonds and binds extracellular components such as collagen fibrin and syndecans.
