Background: FosB/G0S3

The v-Fos oncogene was initially identified as the transforming gene of two independent murine osteosarcoma virus isolates and an avian nephroblastoma virus. The cellular homolog, c-Fos, encodes a 62 kDa nuclear phosphoprotein that is rapidly and transiently induced by a variety of agents and functions as a transcriptional regulator for several genes. In contrast to c-Jun proteins, which form homo and heterodimers, which bind to specific DNA TPA response elements (TREs), c-Fos proteins are only active as heterodimers with members of the Jun gene family. Murine FosB encodes a nuclear protein of 338 amino acids which has 70% homology with c-Fos, exhibits similar kinetics of expression as c-Fos and forms heterodimers with both c-Jun and Jun B which bind to TRE DNA response elements. Functional homologs of c-Fos and FosB include Fra-1 and Fra-2 genes.

Protocols

Western Blot Protocol

1. Perform SDS-PAGE on samples to be analyzed, loading 10-25 ug of total protein per lane.
2. Transfer proteins to PVDF membrane according to the instructions provided by the manufacturer of the membrane and transfer apparatus.
3. Stain the membrane with Ponceau S (or similar product) to assess transfer success, and mark molecular weight standards where appropriate.
4. Rinse the blot TBS -0.05% Tween 20 (TBST).
5. Block the membrane in 5% Non-fat milk in TBST (blocking buffer) for at least 1 hour.
6. Wash the membrane in TBST three times for 10 minutes each.
7. Dilute primary antibody in blocking buffer and incubate overnight at 4C with gentle rocking.
8. Wash the membrane in TBST three times for 10 minutes each.
9. Incubate the membrane in diluted HRP conjugated secondary antibody in blocking buffer (as per manufacturer's instructions) for 1 hour at room temperature.
10. Wash the blot in TBST three times for 10 minutes each (this step can be repeated as required to reduce background).
11. Apply the detection reagent of choice in accordance with the manufacturer's instructions.

  • Q: We are interested in your product, NB100-56530 FosB/G0S3 Antibody, and would like know if this item cross react to mouse. Have you done any in-house test for mouse especially on brain tissue sample. If yes, could you kindly provide some testing result?

    A:

    This antibody was generated by immunizing mice with a synthetic peptide corresponding to amino acids 2-15 of human FosB. This immunogen sequence is 100% analogous to the mouse variant (https://www.uniprot.org/uniprot/P13346), so NB100-56530 will very likely cross react to the mouse protein. Because we have only validated this AB for use in human, we have not done any in-house tests for mouse and don't have any material to offer you. We cannot guarantee that this AB will work for mouse FosB/G0S3, but there is a strong likelihood that it will. 

    Please note that this antibody is hosted in mouse though, which could potentially affect your results since you will need to use an anti-mouse secondary (which in theory will react with the endogenous mouse IgGs in the sample itself). However, since we have not tested this explicitly in mouse, we do not know what the results would look like.
