Q: Could you explain why your G-alpha 13 antibody detects a band at 56kDa while many of the other commercially available antibodies show a band at 44kDa?
A:
The migration of the protein depends on the experimental blot conditions, on the pretreatment of the tested protein samples, as well as on the choice of tested lysates. According to UniProt, the GNA13 protein has several post translational modifications, such as a phosphorylation on Thr-203 and a palmitoylation. Since this protein is said to be a "lipidprotein, a palmitate and a phosphoprotein", I do not think it is surprising that the band may appear larger than expected molecular weight.The link can be found here UniProt Q14344 (https://www.uniprot.org/uniprotkb/Q14344/entry).
