Simple Western: Glucose 6 Phosphate Dehydrogenase Antibody (2H7) - BSA Free [NBP2-22125] - B[a]P-Induced Changes in Cell Cycle. (a) Beas-2B cells treated with 1 µM or 10 µM B[a]P (24 h), 3 µg/mL aphidicolin (18 h), & 100 ng/mL nocodazole (18 h) were prepared for cell imaging post-treatment & immunofluorescent staining. A reference cell cycle diagram is shown. The categorization schematic applied to immunofluorescent images is presented as well as a reference diagram of mammalian cell cycle. Scale bar = 25 μm. (b) Beas-2B cells treated with B[a]P were assessed for WST1 metabolism. n = 5. (c) Quantitative EdU uptake among EdU + Beas-2B cells treated with B[a]P. n = 4. (d) Cumulative cell cycle phase distributions of Beas-2B cells treated with B[a]P or cell cycle controls are presented. n = 4, except nocodazole-treated Beas-2B for which n = 2. (e) Magnitude changes compared to untreated controls among each categorical cell cycle phase are presented; bar colors coincide with phase designations presented in (d). n = 4, except nocodazole-treated Beas-2B for which n = 2. Positive changes denote increased percentage of cells in a specific phase among treated cells, whilst negative values denote decreased percentage. Endpoints are the arithmetic mean of listed independent experiments with error bars denoting one standard error of the mean.
