Specificity

NB100-66437 is specific for gp120, an exterior envelope glycoprotein on the surface of the virus particle that acts as a receptor for CD4 and chemokine receptors on macrophages and T-lymphocytes. It does not react with human T cells, B cells, normal bovine serum or normal human serum. In western blotting a predominant gp120 band is observed.

Background: HIV-1 gp120

HIV1 performs highly complex orchestrated tasks during the assembly, budding, maturation and infection stages of the viral replication cycle. During viral assembly, the proteins form membrane associations and self associations that ultimately result in budding of an immature virion from the infected cell. Gag precursors also function during viral assembly to selectively bind and package two plus strands of genomic RNA. Capsid protein p24 probably forms the conical core of the virus that encapsulates the genomic RNA-nucleocapsid complex.

    A: For IHC, the optimal dilution will have to be determined, as samples can vary significantly. For this antibody a starting dilution of 1:100-1:200 should suffice and then it will have to be determined from there.
