Western Blot Detection of Human DGK-a antibody by Western Blot. View Larger

Detection of Human DGK-alpha by Western Blot. Western blot shows lysates of A431 human epithelial carcinoma cell line and BJAB human Burkitt's lymphoma cell line. PVDF Membrane was probed with 1 µg/mL of Human DGK-a Monoclonal Antibody (Catalog # MAB6597) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). A specific band was detected for DGK-a at approximately 80 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: DGK-alpha

DGK-alpha (Diacylglycerol kinase alpha) is an 80 kDa, 735 amino acid (aa), type I member of the eukaryocytic diacylglycerol kinase family of enzymes, possessing EF‑hand, C1/Cys-rich zinc finger and catalytic domains. In T cells, IL-2 stimulates its translocation to the nucleus, promoting proliferation. TCR stimulation, however, promotes migration to the plasma membrane to downregulate RAS activation and maintain anergy.  It also participates in VEGF or HGF-mediated motility in endothelial, epithelial, or cancer cells. Over aa 1-162, which includes the first of two EF‑hand motifs, human DGK-a shares ~75% aa identity with mouse and rat DGK‑ alpha.
