Background: Cathepsin L
The pro form of insect cell Cathepsin L is stable at neutral pH, but autoactivates in acidic buffers under reducing conditions. The activated enzyme is similar in substrate specificity to mammalian Cathepsin L, and is very active as a protease, as judged by its ability to degrade azocasein. Cystatins A, B, C, E/M, F, SA, and SN are potent inhibitors of the insect cell Cathepsin L. Like other cysteine proteases related to papain, insect cell Cathepsin L is also inhibited by E-64 and reagents which react with thiols.
