Background: Integrin alpha 4/CD49d

VLA-4 belongs to the beta1-integrin family, which functions both as cell-matrix and cell-cell receptors. (1-3) As a matrix receptor VLA-4 binds to an alternatively spliced domain of fibronectin, while as a cellular receptor it associates with its counter-receptor ligand VCAM-1. (1, 2) In addition to CD44/pgp-1, VLA-4 has been shown to be important for lymphopoiesis, and is also believed to be important for extravasation of lymphocytes into sites of inflammation as a second signal in addition to LFA/ICAM-1. (2, 3) CD49d/VLA-4 is expressed in low amounts on resting lymphocytes and monocytes, is absent or minimally detectable on stromal cells, whereas virtually all lymphoid and myeloid cells in bone marrow and in long-term culture are positive. (1, 2) Antibody PS/2 also reacts with human VLA-4. (1)

    Lymph node cells from EAE mice stained for viable cells, VLA4 and CD4. CD4+, VLA4 non-stained cells (red) and CD4+, VLA-stained cells (blue).
    Experimental autoimmune encephalomyelitis was induced in C57BL6/J mice, and periaortic and inguinal lymph node cells were isolated when the animals were at the onset of symptoms. Cells were stained for VLA4, CD4, CD45, LFA-1, TCRb, CD28, CTLA-4, CD40L, OX40, PD1 and CD11b, plus for viability to exclude dead cells. VLA4 staining is showed in CD4+ cells from a control sample not stained for VLA4 (red) and an experimental sample stained for VLA.
