The membrane was probed with 2.0 ug/ml anti-LOXL3 in block buffer and detected with an anti-rabbit HRP secondary antibody using West Pico PLUS chemiluminescence detection reagent.

Immunocytochemistry/Immunofluorescence: Lysyl Oxidase Homolog 3/LOXL3 Antibody [NBP2-75964] - A431 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X PBS + 0.5% Triton-X100. The cells were incubated with anti-LOXL3 at 10 ug/ml overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Background: Lysyl Oxidase Homolog 3/LOXL3

Lysyl oxidase-like protein 3 (LOXL3) is a secreted copper amine oxidase. The N-terminal region of LOXL3 contains four scavenger receptor cysteine-rich (SRCR) domains. The C-terminal region consists of a catalytic domain similar to other lysyl oxidases and a cytokine receptor-like domain. The catalytic domain contains conserved residues required for copper binding and formation of a lysyl tyrosylquinone cofactor. LOXL3 is expressed primarily in epidermis, kidney, aorta, and chondrocytes. The enzyme is expressed at low levels in most other tissues. Like lysyl oxidase, LOXL3 oxidizes the lysine e-amino groups of a number of collagen substrates, and is also active against other primary amines. However, LOXL3 does not require proteolytic processing to be activated.
