Background: Melanocortin-3 R/MC3R

Melanocortins are ligands for a subgroup of GPCRs positively coupled to Adenylate cylcases, termed as melanocortin receptors (MC1R, MC2R, MC3R, MC4R amd MC5R). These receptors are widely expressed at varied density throughout the body. The MC3R has been widely implicated in inflammation and is also implicated in a role in obesity and metabolism. MC3R KO mice exhibit reduced linear growth and lean mass and are hyperleptinemic and hyperinsulinemic.

    Figure 1 shows Lymphocytes stained with Alexa Fluor 488 Rabbit anti-Goat IgG alone, and Figure 2 shows Lymphocytes stained with human MC3R and Alexa Fluor 488 Rabbit anti-Goat IgG.
    Blocking: Natural Mouse Serum Primary antibody: 4 uL (2 ug) Secondary antibody: Alexa Fluor 488 Rabbit anti-Goat IgG (2.5 uL /.5 ug) Details: Human lymphocytes were isolated from PBMCs. Lymphocytes were blocked with Natural Mouse Serum for 15 minutes, and then stained and subsequently incubated at -20 C with anti-human MC3R primary antibody for 30 minutes. After washing, cells were stained with Alexa Fluor 488 Rabbit anti-Goat IgG and incubated at -20 C for 40 minutes. After washing, cells were detected using a Fortessa 4-15 flow cytometer.
