Western Blot Shows Netrin-1 Specificity Using Knockout Cell Line.
Western blot shows cell culture media from MCF7 parental cell line and Netrin-1 knockout MCF7 cell line (KO). Nitrocellulose membrane was probed with Netrin-1 Antibody (SR2201) (Catalog # NBP3-21882) followed by HRP-conjugated secondary antibody. A specific band was detected for Netrin-1 at approximately 67.7 kDa (as indicated) in the parental MCF7 cell line, but is not detectable in knockout MCF7 cell line. Primary antibody dilution used: 1/1000. The Ponceau stained transfer of the blot is shown. This experiment was conducted under reducing conditions. MCF7 cell line cell culture media were prepared and immunoprecipitation was performed using 2.0 μg of Netrin-1 Antibody (SR2201) (Catalog # NBP3-21882) pre-coupled to Dynabeads Protein A. Immunoprecipitated Netrin-1 was detected in Western Blot with a rabbit Netrin-1 antibody used at 1/2000. The Ponceau stained transfer of the blot is shown.
