Immunoblotting analysis of mitophagy proteins. (A) Representative immunoblots for control HT22 cells and mTau-HT22 cells with or without Rlip overexpression and silencing. (B) Represents quantitative densitometry analysis of mitophagy proteins. Pink 1 and Parkin levels were significantly decreased in Mutant Tau when compared with the control. Increased Pink 1 and Parkin levels were observed in Rlip-cDNA with control cells. Decreased Pink 1 and Parkin levels were observed in Tau + Rlip (siRNA) when compared with control HT22 cells. However, significantly increased Pink and Parkin levels were observed in mutant Tau + Rlip-cDNA when compared with the control. (* p = 0.05, ** p = 0.005, *** p = 0.001, and **** p = 0.0001). Application: Western BlotSample Tested: Mouse Neuro2A whole cell lysateSpecies: MouseVerified Customer | Posted 01/22/202135 µg N2A whole cell lysate treated wth 7 different mitochondrial stressors, which should result in parkin stabilization according to the literature. The expected size would be around 55 kDa, which is the fifth band from the top.Neuro2A cells treated witrh different mitochondrial stressors, which should result in parkin stabilization. However I wasn't able to detect parkin, which should run about the same size as the 5th band from the top (55 kDa.) I repeated the experiment and separated again 35 µg whole cell lysate by a 12% PAGE. I labeled the membrane with parkin 1:400 in 5% dry milk powder in TBST. But again nothing. Happy to share that blot as well by email.
