Western Blot: Parkin Antibody (PRK8) [NBP2-29838]

Western Blot: Parkin Antibody (PRK8) [NBP2-29838] - HEK293 cell lysate was probed with anti- Parkin, clone PRK8 (1:2000 dilution). Arrow indicates Parkin (~52kDa).
Parkin Antibody (PRK8)

Western Blot: Parkin Antibody (PRK8) [NBP2-29838] -

Immunoblotting analysis of mitophagy proteins. (A) Representative immunoblots for control HT22 cells and mTau-HT22 cells with or without Rlip overexpression and silencing. (B) Represents quantitative densitometry analysis of mitophagy proteins. Pink 1 and Parkin levels were significantly decreased in Mutant Tau when compared with the control. Increased Pink 1 and Parkin levels were observed in Rlip-cDNA with control cells. Decreased Pink 1 and Parkin levels were observed in Tau + Rlip (siRNA) when compared with control HT22 cells. However, significantly increased Pink and Parkin levels were observed in mutant Tau + Rlip-cDNA when compared with the control. (* p = 0.05, ** p = 0.005, *** p = 0.001, and **** p = 0.0001). Immunoblotting analysis of mitochondrial mitophagy and synaptic proteins. (A) Representative immunoblots for untreated and hAbKI mice and treated hAbKI Urolithin A and EGCG. (B) Quantitative-densitometry analysis of PINK1, Parkin, synaptophysin and PSD95, which shows significant reduction in the hAbKI mice compared to the treated hAbKI mice Urolithin A and EGCG. Background: Parkin

Parkinson's disease is a common neurodegenerative disease with complex clinical features. Mutations in the gene, Parkin (PARK2), appear to be responsible for the pathogenesis of autosomal recessive juvenile Parkinsonism. Parkin plays a role in the ubiquitin-mediated proteolytic pathway by removal and/or detoxification of abnormally folded or damaged protein. Loss of this ubiquitin ligase activity appears to be the mechanism underlying pathogenesis of Parkin. Parkin may protect neurons against alpha synuclein toxicity, proteasomal dysfunction, gpr37 accumulation, and kainate-induced excitotoxicity. It may play a role in controlling neurotransmitter trafficking at the presynaptic terminal and in calcium-dependent exocytosis. Parkin also regulates cyclin e during neuronal apoptosis and may represent a tumor suppressor gene.

35 µg N2A whole cell lysate treated wth 7 different mitochondrial stressors, which should result in parkin stabilization according to the literature. The expected size would be around 55 kDa, which is the fifth band from the top.
Neuro2A cells treated witrh different mitochondrial stressors, which should result in parkin stabilization. However I wasn't able to detect parkin, which should run about the same size as the 5th band from the top (55 kDa.) I repeated the experiment and separated again 35 µg whole cell lysate by a 12% PAGE. I labeled the membrane with parkin 1:400 in 5% dry milk powder in TBST. But again nothing. Happy to share that blot as well by email.
