Immunogen
A synthetic peptide corresponding to a region of the caspase-3-induced cleavage site of human Rb was used as immunogen.
Specificity
The antibody recognizes cleaved Rb (DRb-p70). The antibody does not recognize full-length Rb. In camptothecin treated HL60, an ~68 kDa band is observed. DRb-p70 exists in vivo as an N-terminally truncated form of Rb (Liu et al.). DRb-p70 is thought to be the product of alternative translation and is expressed in primary myeloid cells in fetal liver, bone marrow and spleen. DRb-p70 is also specifically expressed in tumor-associated macrophages (TAMs). TAMs are an important targets of cancer therapy. DRb-p70 is a marker of TAMs, and maybe useful for identifying tumors with macrophage infiltration (Liu et al, 2005).

Western Blot: RB1 Antibody (172C1094) - (cleaved) [NB100-56598] - Western blot shows lysates of HeLa human cervical epithelial carcinoma parental cell line and RB-1 knockout (KO) HeLa cell line. PVDF membrane was probed with 2 ug/ml of Mouse Anti-Human RB-1 Monoclonal Antibody (Catalog # NB100-56598) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog #HAF018). Specific band was detected for RB-1 at approximately 60 kDa (as indicated) in the parental HeLa cell line, but is not detectable in the knockout HeLa cell line.
