PVDF membrane was probed with 0.5 ug/ml of Rabbit Anti-Human MASPIN/Serpin B5 Polyclonal Antibody (Catalog # NBP1-87778) followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog #HAF008). Specific band was detected for MASPIN/Serpin B5 at approximately 40 kDa (as indicated) in the parental HeLa cell line, but is not detectable in the knockout HeLa cell line. This experiment was conducted under reducing conditions.

Background: Serpin B5/Maspin

Maspin is an extracellular matrix tumor supressor that blocks the growth, invasion, and metastatic properties of mammary and prostate tumors. Structurally Maspin is a serine protease inhibitor (Serpin), but it does not undergo the stressed to relaxed conformational transition characteristic of active serpins and exhibits no serine protease inhibitory activity. Maspin is also expressed by the corneal epithelium and regulates migration and adhesion of corneal stromal fibroblasts and myofibroblasts during corneal wound healing.
