Flow Cytometry: SMAC/Diablo Antibody (006) [NBP2-89251] - Flow cytometric analysis of Human SMAC/Diablo expression on HeLa cells. The cells were treated, stained with NBP2-89251 (Filled histogram), then a FITC-conjugated second step antibody. To demonstrate specificity of staining, the binding by NBP2-89251 was blocked by preincubation of the purified antibody with 20ug recombinant human SMAC/Diablo for 1 hour (Black solid line hisgram). The fluorescence histograms were derived from gated events with the forward and side light-scatter characteristics of intact cells.

Immunocytochemistry/ Immunofluorescence: SMAC/Diablo Antibody (006) [NBP2-89251] - Immunofluorescence staining of Human SMAC/Diablo in Hela cells. Cells were fixed with 4% PFA, permeabilzed with 0.3% Triton X-100 in PBS, blocked with 10% serum, and incubated with NBP2-89251 (1:60) at 4 overnight. Then cells were stained with the Alexa Fluor 488-conjugated Goat Anti-rabbit IgG secondary antibody (green) and counterstained with DAPI (blue). Positive staining was localized to cytoplasm.

Background: SMAC/Diablo

Smac/DIABLO is a mitochondrial protein which activates various forms of apoptosis. Specifically, Smac exits the mitochondria and enters the cytosol where it initiates apoptosis by neutralizing members of the IAP family of apoptosis inhibitory proteins. Mitochondrial-mediated apoptosis is important in animal development and tissue homeostasis, with alterations resulting in a range of malignant disorders. Upon apoptotic stimuli, the mitochondrial proteins cytochrome c and Smac/DIABLO are released into the cytosol. The release of these proteins, however, occurs via different mechanisms.
