Background: VAP-1/AOC3

The monoclonal antibody 174-5 recognises human Vascular Adhesion Protein-1 (VAP-1) also known as AOC3, a heavily sialylated homodimeric glycoprotein of 180 kD. VAP-1 is present in endothelial cells, smooth muscle cells, adipocytes, and in follicular dendritic cells. The majority of VAP-1 is stored within intracellular granules and translocated to the endothelial surface on inflammation. Structurally it belongs to enzymes called semicarbamizide-sensitive amine oxidases. These enzymes deaminate primary amines in a reaction producing hydrogen peroxide, aldehyde, and ammonia. VAP-1 possesses both adhesive and enzymatic properties in vitro. VAP-1 mediates leukocyte trafficking to sites of inflammation. The monoclonal antibody 174-5 cross-reacts with rat VAP-1. The monoclonal antibody inhibits lymphocyte infiltration in liver allograft rejection in rats.

  • Q: One of our customers is very interested in your Antibody: VAP1 Antibody (174-5). This antibody is not listed as functional assays, but in the reference cited in the data sheet, there is an article referring this antibody as blocking VAP-1 in vivo in rats. The article is: Martelius, T et al. Blockade of vascular adhesion protein-1 inhibits lymphocyte infiltration in rat liver allograft rejection. Am J Path 2004, 165: 199. Could you please confirm that this antibody could be used in Functional assays? Also the format is really small and the customer need huge quantity of antibody as the recommended dosys is 2mg/kg. Do you have any other bigger format?

    A: This antibody has not been tested in functional assays. It has been validated for use in Flow Cytometry, Western Blot, Immunofluorescence and Immunohistochemistry-Frozen. The reference you refer to has been taken off our website. I apologize for any confusion.
