Immunocytochemistry: WT1 Antibody (960522) [MAB5729] - WT-1 was detected in immersion fixed K-562 cell line using MAB5729 at 8 ug/ml for 3 hours at room temperature. Cells were stained using NL007 (red) and counterstained with DAPI (blue). Specific staining was localized to cytoplam & nuclei.

Flow Cytometry: WT1 Antibody (960522) [MAB5729] - An intracellular stain was performed on THP-1 cells with WT1 [960522] Antibody MAB5729AF647 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 2.5 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to Alexa Fluor 647.

Background: WT1

Wilms' tumor protein (WT1) is a transcription factor which regulates the expression of numerous target genes, including EPO. This protein plays an essential role in the normal development of the urogenital system, including cellular development and cell survival, and it has both tumor suppressor and oncogenic roles in tumor formation.

Defects in the WT1 protein can lead to Wilms tumor, Frasier syndrome, Denys-Drash syndrome, nephrotic syndrome type 4, and Meacham syndrome.
