Comet assay, also known as single cell gel electrophoresis assay, is a powerful technique for evaluating DNA damage and repair. The comet assay technique utilizes cells that are embedded into agarose, placed on a slide, and then immersed in lysis solution. After cells have been lysed, DNA is denatured and run through an electrophoresis. The data image that is obtained resembles that of a comet which includes a distinct comet head and comet tail. The head of the comet contains DNA that is still intact while the comet tail consists of the damaged DNA. Quantitative and statistical data do the comets can be generated by analysis of the results using the CometAssay Analysis Software, which can calculate tail length, percent DNA in the tail, and tail moment.

CometChip is a high-throughput platform version of the comet assay that utilizes a 96 well plate with micron pores patterned into agarose. Cells in each well are captured in micropores while excess cells are aspirated. This leaves an array of non-overlapping cells that can then be lysed and run through electrophoresis. CometChip provides researchers the ability to run and analyze hundreds of comet assays in a fraction of the time compared to a standard comet assay.
