Specificity

Detects mouse BMP-5 in direct ELISAs and Western blots. In Western blots, approximately 20% cross-reactivity with recombinant human BMP-5 is observed. In direct ELISAs, approximately 100% cross-reactivity with recombinant human BMP-5 is observed, and approximately 4% cross-reactivity with recombinant mouse (rm) BMP-6 and rmBMP-7 is observed, and less than 1% cross-reactivity with rmBMP-8b is observed.

Product Feature: CoraFluor Probes for TR-FRET

CoraFluor™ 1, amine reactive (Catalog:7920) and CoraFluor™ 2, amine reactive (Catalog # 7950) are terbium-based probes that have been developed for use as TR-FRET donors. They emit wavelengths compatible with commonly used fluorescent acceptor dyes such as BODIPY® (or BDY) and Janelia Fluor® dyes, FITC (Catalog # 5440), TMR and Cyanine 5 (Catalog # 5436). CoraFluor™ fluorescence is brighter and more stable in biological media than existing TR-FRET donors, leading to enhanced sensitivity and improved data generation. CoraFluor™ 1 exhibits excitation upon exposure to a 337 nm UV laser.

Background: BMP-5

Bone morphogenic proteins (BMPs) are members of the TGFbeta superfamily. BMPs are involved in the induction of cartilage and bone formation. In vivo studies have shown that BMP-2 (also designated BMP-2A) and BMP-3 can independently induce cartilage formation. Smad3 association with the TGFbeta receptor complex and Smad1 translocation to the nucleus are observed after the addition of BMP-4 (also designated BMP-2B), suggesting that BMP-4 may play a role in activation of the Smad pathway. BMP-5, BMP-6 and BMP-7 all share high sequence homology with BMP-2, indicating that they each may be able to induce cartilage formation. BMP-8 (also designated OP-2) is thought to be involved in early development, as detectable expression has not been found in adult organs.
