Neutralization

Measured by its ability to neutralize Recombinant Human Serpin A4 (3.8 µg/mL, Catalog # 1669-PI) inhibition of Chymotrypsin (0.1 µg/mL) cleavage of the fluorogenic peptide substrate Mca-RPKPVE-Nval-WRK(Dnp)-NH₂ (10 µM, Catalog # ES002). The Neutralization Dose (ND₅₀) is typically 10.8 µg/mL.

Detection of Human Serpin A4/Kallistatin by Western Blot. Western blot shows lysates of human liver tissue, human kidney tissue, and human pancreas tissue. PVDF membrane was probed with 0.25 µg/mL of Goat Anti-Human Serpin A4/Kallistatin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF1669) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF017). Specific bands were detected for Serpin A4/Kallistatin at approximately 50-60 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: Serpin A4/Kallistatin

Serpin A4 is a member of the Serpin superfamily of the serine protease inhibitors (1). Known also as Kallistatin or proteinase inhibitor 4, Serpin A4 has been shown to function as a specific inhibitor of tissue kallikrein and as an inhibitor of angiogenesis and tumor growth (2, 3).
