Immunogen

Derived by fusion of NS0 Mouse myeloma cells with spleen cells from a BALB/c Mouse immunized with a peptide comprising the N-terminal nonapeptide of gamma-cytoplasmic actin with an acetylated N-terminus (Ac-EEEIAALVI-COOH) coupled to keyhole limpet hemocyanin through the cysteine residue. The antibody does not cross react with other actin isoforms.

Reactivity Notes

The epitope recognized is highly conserved. The antibody therefore cross-reacts with many species including protochordates, lower craniates and mammals. Please note that this antibody is reactive to Mouse and derived from the same host, Mouse. Mouse-On-Mouse blocking reagent may be needed for IHC and ICC experiments to reduce high background signal. You can find these reagents under catalog numbers PK-2200-NB and MP-2400-NB. Please contact Technical Support if you have any questions.

Specificity

This mouse monoclonal IgG2b antibody highly specific for gamma-cytoplasmic actin (Actin Gamma 1; encoded by ACTG1). It does not cross-react with the Actin Gamma 1 isoform expressed in the enteric smooth muscle tissues (actin gamma 2; encoded by ACTG2).

Background: Actin Gamma 1

Actin, gamma 1 (ACTG1) is a member of the actin protein family, a highly conserved protein group with three main isoforms: alpha, beta and gamma. Alpha actins are mainly expressed in muscle tissues where they are a major constituent of the contractile apparatus, whereas beta and gamma actin proteins are widely expressed in most cell types where they are predominantly involved in cell motility and cytoskeleton maintenance. Actin gamma 1 is a cytoplasmic actin isoform usually found in nonmuscle cells.

Actin Gamma 1 antibodies serve as excellent loading controls, due to the proteins' near ubiquitous expressn and strong conservation across most species. In vertebrates 3 main groups of actin isoforms, alpha, beta and gamma have been identified. The alpha actins are found in muscle tissues and are a major constituent of the contractile apparatus. The beta and gamma actins coexist in most cell types as components of the cyto-skeleton and as mediators of internal cell motility. Beta actins are highly conserved proteins that are involved in cell motility, structure and integrity. Beta actins are cytoplasmic proteins ubiquitously expressed in all eukaryotic cells. Polymerization of globular actin (G-actin) leads to a structural filament (F-actin) in the form of a two-stranded helix. Each actin can bind to 4 others. Actin Gamma 1 antibody serves as an excellent loading control.
