Application Notes
ELISA confirmed the specificity of 2A11 for the V-domain of AGER, with a binding affinity (Kd) of 7.6 nM. ELISA was also used to confirm that the binding of 2A11 to AGER is glycosylation independent. In SPR experiments the Kd of 2A11 binding to AGER was determined as 0.38 nM. Flow cytometry and immunochemistry were used to visualise the binding of 2A11 to HEK-AGER cells.

AGER encodes a member of the immunoglobulin superfamily of cell surface molecules. It is a receptor for various molecules, including the amyloidogenic form of serum amyloid A, amyloid-beta protein, members of the S100/calgranulin superfamily and advanced glycation end products. The gene lies within the major histocompatibility complex (MHC) class III region on chromosome 6. Alternative splicing results in two transcript variants encoding different isoforms.
