Specificity
The Apoptosis Detection Antbody Bundle contains monoclonal antibodies to three key apoptosis markers: caspase-3, caspase-8, and cleaved PARP. The caspase-3 and -8 antibodies both recognize pro and cleaved forms. The PARP antibody specifically recognizes the 89 kDa PARP fragment and does not recognized the full-length protein.

Western Blot: Apoptosis Detection Antibody Pack [NBP2-25080] - Image of anti-Caspase 3 (31A1067). Whole cell protein from Jurkat cells treated with and without 2 uM staurosporine as indicated was separated on a 4-15% gel by SDS-PAGE, transferred to 0.2 um PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 5 ug/ml anti-Caspase 3 (NB100-56708) in 1% milk, and detected with an anti-mouse HRP secondary antibody using a Femto sensitivity chemiluminescence reagent. Note the detection of both pro-caspase 3 at 35 kDa and the cleaved active caspase 3 at 15-17 kDa.Western Blot: Apoptosis Detection Antibody Pack [NBP2-25080]
Western Blot: Apoptosis Detection Antibody Pack [NBP2-25080] - Analysis of Caspase-8 in Jurkat cells using at 1 ug/ml. Cells were treated with 2 uM staurosporine for different time periods. Caspase-8 activation is detected in western blot by the presence of Caspase-8 cleavage fragments. The antibody detected both pro (full-length) and active (cleaved) Caspase-8, depending on the treatment time points. A basal level of endogenously cleaved Caspase-8 can be see in untreated Jurkat cells. this antibody was used in this test.Western Blot: Apoptosis Detection Antibody Pack [NBP2-25080]
Western Blot: Apoptosis Detection Antibody Pack [NBP2-25080] - Analysis of cleaved PARP in staurosporin-treated Jurkat cells at various time points, using at 2 ug/ml.
