Western Blot: ATF4 Antibody (SD20-92) [NBP2-67766] - Analysis of ATF4 on different lysates. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in PBS for 1 hour at room temperature. The primary antibody (1:500) was used in 5% BSA at room temperature for 2 hours. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1:5,000 dilution was used for 1 hour at room temperature. Tyrosine hydroxylase chicken in red and ATF4 in green No specific signal for ATF4 at 1/50emeMice have been deeply anesthetized and perfused with cold PBS and then PFA. Brains have been dissected and fixed in PFA 4% overnight at 4°C. Coronal sections of substantia nigra of 40 µm thickness have been cut using a cryostat. For immunostaining, free-floating sections have been incubated in TBS with bovine serum albumin, triton, goat serum during 1h. Then primary antibodies during 2h and we have used secondary antibodies Goat anti rabbit for ATF4 in 488 1h time incubation.
