S@A-NPs treatment suppresses amyloidogenic cleavage of APP, while enhancing transcytosis of A beta out of the brains of APP/PS1 transgenic mice. (a) Schematic description of experimental timeline. APP/PS1 mice were intravenously injected with S@A-NPs at 6 months of age and subjected to analysis of A beta clearance at 7 months of age. SF555-conjugated A beta oligomers were stereotactically injected into the hippocampal DG. (b, c) SF555-conjugated A beta remnants in the brains were shown 48 h after injection. Area of SF555-conjugated A beta oligomers occupied in the hippocampal DG (c). (d, e) ELISA analysis of A beta 42 (d) and A beta 40 (e) levels in the plasma. (f) WB analysis of levels of full-length APP, BACE1 and alpha / beta -CTF in the cortex and hippocampus. GAPDH was detected as a loading control. Relative levels of full-length APP (f, g), BACE1 (f, h), beta -CTF (f, i), and alpha -CTF (f, j). Data are presented as mean +/- SEM. n = 3 mice/group (c); n = 7–9 mice/group (d); n = 6–11 mice/group (e); n = 3–8 mice/group (g-j). * p < 0.05; ** p < 0.01; *** p < 0.001. One-way ANOVA (c-e, g-j). Background: BACE-1

BACE1 is a protease responsible for the proteolytic processing of the amyloid precursor protein (APP). Amyloid beta peptide is the major constituent of amyloid plaques in the brains of individuals afflicted with Alzheimers disease. This peptide is generated from the beta-amyloid precursor protein (beta APP) in a two-step process. The first step involves cleavage of the extracellular, amino-terminal domain of beta APP. Protein cleavage is performed by an aspartyl protease termed beta-secretase (BACE), of which there are two isoforms, BACE1 and 2. This enzyme is synthesized as a propeptide that must be modified to the mature and active form by the prohormone convertase furin. Beta APP cleavage by the mature form of BACE results in the cellular secretion of a segment of beta APP and a membrane-bound remnant. This remnant is then processed by another protease termed gamma-secretase. Gamma-secretase cleaves an intra-membrane site in the carboxyl-terminal domain of beta APP, thus generating the amyloid beta peptide.
