After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-rabbit polymer IgG labeled by HRP and visualized using 0.05% DAB.

Immunocytochemistry/Immunofluorescence: BCAP31 Antibody (12G12) [NBP3-26481] - Staining of A549 cell with BCAP31 Antibody (12G12) at 1:50, counter-stained with DAPI. The cells were fixed in 4% formaldehyde and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4C. The secondary antibody was Alexa Fluor 526-conjugated Goat Anti-Rabbit IgG (H+L).

Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1ug/1*10^6 cells) for 45min at 4C. The secondary antibody used was FITC-conjugated Goat Anti-rabbit IgG (H+L) at 1:200 dilution for 35min at 4C. Control antibody (green line) was rabbit IgG (1ug/1*10^6 cells) used under the same conditions. Acquisition of >10,000 events was performed.

Background: BAP31

BAP31 (B Cell Receptor Associated Protein, 28 kDa) is a multiple membrane spanning protein of the endoplasmic reticulum. It has been shown to form both homo and hetero oligomers with the closely related BAP29, and is a potential apoptotic regulator as a predicted BCL2/BCLXL associated protein. In the C terminal domain of BAP31, there are two identical recognition sites for initiator Caspases 1 and 8 of the programmed death cascade. It is hypothesized that BAP31 plays a role in the transport of selected proteins to the Golgi apparatus. BAP31 has also been shown to control expression of CFTR (cystic fibrosis transmembrane conductance regulator). It is believed that interference with the expression or function of BAP31 in epithelial cells may be a way to circumvent the chloride channel defect in cystic fibrosis.
